چکیده
Monoclonal antibodies targeting tumors are one of the most important discoveries in field cancer. Although several effective have been developed, a relapse may occur. One their mechanisms action is Antibody Dependent Cell Cytotoxicity (ADCC), by engaging Fc γ receptor CD16 expressing Natural Killer cells, innate lymphoid cells involved cancer immunosurveillance and able to kill tumor cells. A lack NK observed many cancers therefore be cause low efficacy some clinical situations. Here we review clear evidences essential partnership between showed vitro, vivo, trials different indications, describe hurdles ways enhance ADCC evolution monoclonal antibody therapy. cell adoptive immunotherapy combined with overcome resistance treatment efficacy.

Natural Killer Cell Mediated Antibody-Dependent Cellular Cytotoxicity in Tumor Immunotherapy with Therapeutic Antibodies
In the last decade several therapeutic antibodies have been Federal Drug Administration (FDA) and European Medicines Agency (EMEA) approved. Although their mechanisms of action in vivo is not fully elucidated, antibody-dependent cellular cytotoxicity (ADCC) mediated by natural killer (NK) cells is presumed to be a key effector function. A substantial role of ADCC has been demonstrated in vitro ...
متن کاملCytotoxicity of activated natural killer cells against pediatric solid tumors.
PURPOSE To develop new therapies for children with solid tumors, we tested the cytotoxicity of natural killer (NK) cells expanded by coculture with K562-mb15-41BBL cells. We sought to identify the most sensitive tumor subtypes, clarify the molecular interactions regulating cytotoxicity, and determine NK antitumor potential in vivo. EXPERIMENTAL DESIGN We tested in vitro cytotoxicity of expand...
متن کاملThe role of natural killer cells and antibody-dependent cell-mediated cytotoxicity during murine cytomegalovirus infection
Definition of the functions by which the cellular immune system contributes to control of cytomegalovirus (CMV) infection should permit determination of the specific defects which result in the increased susceptibility to infection of immunosuppressed individuals. Using a murine model, we studied the cytotoxic lymphocyte response to murine CMV infection. This response was found to be biphasic. ...
متن کاملlenalidomide enhances natural killer cell and monocyte-mediated antibody-dependent cellular cytotoxicity of rituximab-treated CD20+ tumor cells.
PURPOSE Lenalidomide has significant activity in myelodysplastic syndromes, multiple myeloma, and non-Hodgkin's lymphoma (NHL). In previous studies, natural killer (NK) cell expansion by lenalidomide was shown to enhance the cytotoxic effect of rituximab. This study assessed the ability of lenalidomide to enhance antibody-dependent cellular cytotoxicity (ADCC) in rituximab-treated NHL cell line...
متن کاملAntibody-dependent cellular cytotoxicity toward neuroblastoma enhanced by activated invariant natural killer T cells.
Anti-ganglioside GD2 antibodies mainly work through antibody-dependent cellular cytotoxicity (ADCC) and have demonstrated clinical benefit for children with neuroblastoma. However, high-risk neuroblastoma still has a high recurrence rate. For further improvement in patient outcomes, ways to maximize the cytotoxic effects of anti-GD2 therapies with minimal toxicity are required. Activated invari...
